چکیده

Fluorescence correlation spectroscopy (FCS) is a well-established method for the analysis of freely diffusing fluorescent particles in solution. In a two-colour setup, simultaneous detection of two different dyes allows the acquisition of both the autocorrelation of the signal of each channel and the cross-correlation of the two channels (fluorescence cross-correlation spectroscopy, FCCS). The cross-correlation function is related to the amount of diffusing particles carrying both dyes and can be used for monitoring a binding reaction. Here we develop a formalism for a quantitative analysis of ligand binding from a combination of the autoand the cross-correlation amplitudes. Technical constraints, like the focal geometry, background signal and cross-talk between the detection channels as well as photophysical and biochemical effects which modulate the brightness of the particles are included in the analysis. Based on this framework a comprehensive treatment for the determination of two-component binding equilibria by FCS/FCCS is presented.

Measuring Protein Binding to Lipid Vesicles by Fluorescence Cross-Correlation Spectroscopy.

Fluorescence correlation spectroscopy has been previously used to investigate peptide and protein binding to lipid membranes, as it allows for very low amounts of sample, short measurement times and equilibrium binding conditions. Labeling only one of the binding partners, however, comes with certain drawbacks, as it relies on identifying binding events by a change in diffusion coefficient. Sin...

Filtered FCS: Species Auto- and Cross-Correlation Functions Highlight Binding and Dynamics in Biomolecules**

An analysis method of lifetime, polarization and spectrally filtered fluorescence correlation spectroscopy, referred to as filtered FCS (fFCS), is introduced. It uses, but is not limited to, multiparameter fluorescence detection to differentiate between molecular species with respect to their fluorescence lifetime, polarization and spectral information. Like the recently introduced fluorescence...

A spectroscopic study on Calf thymus DNA binding properties of nickel (II) complex with imidazole derivatives of 1,10-phenanthroline ligand

In this study, a nickel (II) complex with 1,10-phenanthroline based ligand, [Ni(FIP)2](OAC)2 (1) with FIP = 2-(Furan-2-yl)-1H-Imidazole[4,5-f][1,10] phenanthroline as ligand was synthesized and characterized by spectroscopic methods and elemental analysis. The interaction of [Ni(FIP)2](OAC)2 (1) with calf-thymus DNA (ct-DNA) was studied by UV-vis absorption, fluorescence spectroscopies and visc...

Dual-color total internal reflection fluorescence cross-correlation spectroscopy.

We present the development and first application of a novel dual-color total internal reflection (TIR) fluorescence system for single-molecule coincidence analysis and fluorescence cross-correlation spectroscopy (FCCS). As a performance analysis, we measured a synthetic DNA-binding assay, demonstrating this dual-color TIR-FCCS approach to be a suitable method for measuring coincidence assays su...

A two-photon excitation fluorescence cross-correlation assay for a model ligand-receptor binding system using quantum dots.

Two-photon excitation fluorescence cross-correlation spectroscopy (TPE-XCS) is a very suitable method for studying interactions of two distinctly labeled fluorescent molecules. As such, it lends itself nicely to the study of ligand-receptor interactions. By labeling the ligand with one color of fluorescent dye and the receptor with another, it is possible to directly monitor ligand binding rath...
